ASSAY Sensitivity
The maximum sensitivity of Chromo-LAL reagent is 0.005 EU/mL. The sensitivity for chromogenic assays is determined by the lowest standard concentration on the standard curve used for the assay.
Sample to Lysate Ratio
Sample-to-Chromo-LAL ratio is 1:1 (100uL : 100uL)
Test Performance
Per validated conditions, the reaction mixture is incubated at 37 ± 1°C in a microplate reader equipped with a 405-410 nm filter. The incubation time depends on the lowest standard concentration and the lot number of the reagent. Software is used to construct the standard curve and calculate the endotoxin concentrations.
Product Stability
The reconstituted lysate remains stable for up to 24 hours when stored at 2–8°C or for two weeks at -20°C or colder if frozen immediately after reconstitution and not contaminated. The reconstituted Chromo-LAL lysate may be frozen and thawed once.
Reconstitution
Chromo-LAL lysate should be reconstituted with LAL Reagent Water (LRW). To render the assay endotoxin-specific, it can also be reconstituted with Glucashield® (1→3)-β-D-glucan inhibiting buffer.
Ordering Information
Chromo-LAL is recommended for use with 0.5 ug/vial Control Standard Endotoxin (CSE, E0005). Certificates of Analysis, specific to the Pyrochrome® and CSE lot, can be obtained on our website . Chromo-LAL is sold as a pack of 5 vials. This allows you to order the amount of lysate that is right for you.
