Product Sensitivity

Maximum sensitivity of Pyrotell®-T is reader dependent:

  • Pyros Kinetix® Flex tube reader:  0.001 EU/mL, which is 5 times more sensitive than most BET assays. 
  • Microplate reader: 0.005 EU/mL.

However, the range of standard curve used should be adapted per user needs reflecting the actual conditions of the test. 

Sample-to-Lysate Ratios

Sample-to-Pyrotell®-T ratio is reader dependent:

  • Pyros Kinetix® Flex tube reader: 1:1 (100ul: 100ul) or 4:1 (200ul : 50ul) 
  • Microplate reader: 1:1 (100ul: 100ul) 

If sample interference is observed at 4:1 ratio, the assay should be repeated at 1:1 ratio.

Each vial of Pyrotell®-T contains 5mL of reagent. The number of tests per vial depends on the volume used per test (either 50 tests/vial or 100test/vial). 

Test Performance

Per validated conditions. The reaction mixture is incubated in 8x75mm borosilicate reaction tubes or a microplate and incubated at 37±1°C and the developing absorbance is read at visible light wavelength (reader dependent). The incubation time depends on the lowest standard concentration and the lot number of the reagent. Software is used to construct the standard curve and calculate the endotoxin concentrations.

Product Stability

Once reconstituted, Pyrotell®-T lysate is stable for 24 hours if stored at 2–8°C. Pyrotell®-T lysate may be frozen once and will retain activity for as long as 3 months if stored at or below -20°C.

Pyrotell®-T is recommended for use with 0.5 ug/vial Control Standard Endotoxin (CSE, E0005). Certificates of Analysis, specific to the Pyrotell®-Tand CSE lot, can be obtained on our website.

Pyrotell®-T is available as a pack of 5 or 25 vials.
